What Each Tier Means

Introductory — 0.1 mg/kg

A conservative starting point used in some community protocols when first introducing methylene blue. At this level, the compound's characteristic blue coloring of urine is minimal and any individual sensitivity can be gauged. It is below the range examined in most published cognitive research but above a purely symbolic dose.

Low Range — 0.5 mg/kg

The lower end of the range most commonly examined in peer-reviewed cognitive and mitochondrial research. For a 70 kg person, this is 35 mg — approximately 35 drops or 1.75 mL of a 2% solution. Rojas et al. (2012) documented that spatial memory performance approximately doubled at doses in this range compared to controls, without the diminishing returns seen at higher doses.

Standard Range — 1 mg/kg

The upper end of the most frequently cited research range and the most common reference point in community protocols. For a 70 kg person, this is 70 mg — approximately 70 drops or 3.5 mL of a 2% solution. The Rodriguez et al. (2016) human randomized controlled trial used approximately 4 mg/kg (280 mg for a 70 kg person) and documented a 7% improvement in memory retrieval, but most community protocols operate in this 1 mg/kg range rather than the higher clinical trial dose.

A Note on Drop Accuracy

The calculator shows drop counts using the standard reference of 1 drop ≈ 1 mg for a 2% (20 mg/mL) solution — because a standard dropper delivers approximately 20 drops per mL, and 1 mL of a 2% solution contains 20 mg, meaning each drop contains roughly 1 mg.

However, drop size is not standardized. It varies with dropper tip diameter, bottle angle, liquid temperature, and surface tension. At doses of 25–100 mg (1.25–5 mL), individual drop variability adds up. A graduated 1 mL oral syringe delivers more consistent and reproducible measurements. The mL column in the calculator is the more reliable number to work from.


Concentration Assumption

This calculator is built for a 2% methylene blue solution (20 mg/mL). If your solution is a different concentration, the drop and mL values will not apply — only the mg values remain valid. Common concentrations and their mg/mL equivalents:

  • 1% solution — 10 mg/mL (double the mL and drops shown)
  • 2% solution — 20 mg/mL (what this calculator assumes)
  • 3% solution — 30 mg/mL (use 2/3 the mL and drops shown)
