Why morning dosing works best
Methylene blue reaches peak blood concentration approximately 1 to 2 hours after oral dosing, with a half-life of 5 to 6.5 hours. This means a morning dose peaks in the mid-morning, when most people are doing their best cognitive work, and clears sufficiently by evening to avoid sleep disruption. A dose taken at noon with a 10 PM bedtime would still have roughly 25% of peak concentration in the blood at sleep onset — enough to affect sleep quality in sensitive individuals.
The 6-hour rule and sleep
The most common timing mistake with methylene blue is dosing too late. Because it inhibits MAO-A (the enzyme that breaks down serotonin and dopamine), late dosing can prolong the time these neurotransmitters remain active, which may delay sleep onset and reduce sleep quality. Most protocols suggest a hard cutoff of 6 hours before your target sleep time. For a 10 PM bedtime, this means the last dose should be at 4 PM at the latest — and ideally before noon.
